Overview

This book reviews knowledge on the interconnection of signal transduction and endocytosis/intracellular trafficking. The chapters cover knowledge obtained by using different model systems. The first chapter deals with Receptor Tyrosin Kinases (RTKs) with emphasis on the Epidermal Growth Factor Receptor (EGF receptor) and the Platelet Derived Growth Factor Receptor (PDGF receptor). The second chapter deals with the RTK c-Met and with how this RTK becomes carcinogenic. The third chapter reviews recent understanding on the mechanisms of action of the numerous fibroblast growth factors and their receptors. In the fourth chapter we learn about the trafficking of and signalling from the Growth Hormone Receptor and how this receptor is controlled by ubiquitination. The fifth chapter is devoted to the Interleukin II receptor, essential for activation of T cells. Links between ubiquitination, signalling, endocytosis, and sorting are reviewed. The last chapter discusses current views on how monoubiquitination controls both signalling and trafficking and thereby the final outcome of receptor activation.

Description: This book, a volume in the series, Current Topics in Microbiology and Immunology, provides a comprehensive review of the biochemistry literature on signaling mechanisms with special emphasis on receptor internalization and ubiquitination. As noted by the editor, receptors are critically important in cellular physiology, because they control growth, apoptosis, differentiation and cancer progression. The six chapters focus on model signaling systems, covering topics ranging from the intracellular activity of fibroblast growth factor and hepatocyte growth factor to signaling through monoubiquitination.
Purpose: The goals of the editor and authors are to summarize key experimental findings that highlight the importance of endocytosis and intracellular processing in cell signaling. The contributions are up-to-date and informative.
Audience: Targeted at research scientists in oncology, biochemistry, pharmacology, and molecular biology, the book is written primarily for specialists working on mechanisms of transmembrane signaling in mammalian cells. The authors are research scientists and experts in their field.
Features: This book assumes a high level of understanding of cellular receptors and intracellular signaling mechanisms. Key model systems illustrate basic principles that appear to govern most receptor-mediated signaling pathways. Each contribution includes a comprehensive list of key references. The book contains many illustrative line drawings, including a few that are reproduced in color. It is compact and readable.
Assessment: Unfortunately, the authors merely summarize the existing literature. They do not critically evaluate the experimental findings. They do not integrate knowledge or formulate new hypotheses. Nonetheless, this book is a very useful summary of the existing literature on signaling mechanisms for experts working in the field.
