Nella famosa abitazione di Baker Street al 221/b giunge un visitatore sconosciuto: è un medico di campagna di nome Mortimer, che per anni ha seguito le vicende di una famiglia nobile che vive a Baskerville, nella contea di Devon. Tale famiglia ha nella sua storia una drammatica vicenda legata alla diceria che un mostro avrebbe ucciso nel Settecento un avo molto crudele. Holmes accetta l'incarico proposto da sir Henry, ovvero scoprire cosa stia avvenendo nel castello dei Baskerville, ma decide di inviare il fidato Watson. Quest'ultimo e sir Henry si recano nella dimora avita dei Baskerville. Da questo momento il romanzo si sposta su un piano colmo di situazioni inaspettate, incubi, misteri, che accrescono i dubbi coinvolgendo personaggi che nascondono storie segrete, forse tutti quanti in un rapporto non troppo chiaro con la vicenda della morte di sir Charles. E Watson indaga mentre il giovane erede sir Henry si sente minacciato. Intanto nella brughiera si odono al tramonto ululati raccapriccianti; inoltre un evaso si è nascosto nella palude e un uomo misterioso sta osservando di nascosto cosa sta accadendo.
