Description: This 15-chapter book is a compilation of topics related to current clinical uses of monoclonal antibodies and their associated protocols.
Purpose: The purpose was to gather together reviews and related laboratory protocols for current clinical uses of monoclonal antibodies. These are worthy goals given the increasing use of monoclonal antibodies as reagents and pharmaceutical agents that the book meets.
Audience: The audience includes those interested in bringing the discussed assays into their own laboratories or in the various clinical applications of monoclonal antibodies today. The editor and many of the authors are credible authorities in this field.
Features: The book has two major sections. The first three chapters provide broad overviews on mouse hybridomas, recombinant monoclonal antibodies, and soluble Fc fusion proteins. The remaining 12 chapters discuss applications of monoclonal antibodies to clinical issues, i.e., assisting diagnoses with monoclonal antibody staining (immunohistochemical or flow cytometry based methods) to measuring various molecules of interest (e.g., FLT3, bcr-abl) in various fluids or tissues. Many of the chapters assume basic knowledge in the topic. For example, at least two chapters are devoted to using monoclonal antibodies for refining the diagnosis of various hematological disorders (e.g., leukemia, lymphoma) with very little discussion of the disorders themselves or the diagnostic process needed to get to the point of using monoclonal antibody reagents. The chapters are uneven in content, with some providing extensive background material and others simply reporting their own applied research (e.g., a chapter discussing detection of native versus phosphoylated bcr-abl protein). A plus is the inclusion of laboratory protocols so the reader can immediately implement an assay. A few detractions and inconsistencies are readily apparent. The final chapter discussing allergen specific IgE consistently misspells basophils as "basophiles," yet the index lists them as basophils. The algorithms for distinguishing between acute myelogenous leukemia and acute lymphocytic leukemia are not provided within the context of the major classification schemes (i.e., WHO classification). One set of immunohistochemical figures are in color with the remainder in black and white; why not all in color? And finally, the sensitivity (analytical and clinical) of monoclonal antibodies as assay reagents is discussed, yet there is no comparable discussion of their specificity (analytical or clinical).
Assessment: This is the book for you if you are interested in implementing one or more of these assays in your laboratory.
